What is IGF-1 DES?

IGF-1 DES is insulin-like growth factor 1 missing its first three amino acids, Gly-Pro-Glu. That leaves a 67-amino-acid protein of about 7.4kDa that still binds the IGF-1 receptor.

It was found naturally in bovine colostrum and later in human brain tissue in the mid-1980s. The missing three residues are exactly where IGF binding proteins grab hold, so very little of it gets tied up.

How it works

IGF-1 works through the IGF-1 receptor, a tyrosine kinase that turns on the PI3K-Akt and MAPK pathways. Those pathways push cells to grow, divide and resist cell death.

In tissue, most IGF-1 is held by six binding proteins that keep it from reaching the receptor. Without its first three residues, IGF-1 DES slips past them, so more of it is free to act locally. It also clears faster than IGF-1 LR3.

Published IGF-1 DES research

Francis, Ballard and colleagues in Adelaide reported that des(1-3)IGF-1 was several times more potent than normal IGF-1 in cell culture, and showed the reason was its weak binding to IGF binding proteins.

That finding led to the design of IGF-1 LR3, which uses the same trick plus an extension to last longer. Most research use is in cell culture and growth studies.

Study design notes

GH-axis studies often look at IGF-1 as the steadier marker, since a single GH reading can miss a pulse. Cortisol and prolactin are worth measuring too, because some secretagogues raise them.

Run each peptide on its own before you test combinations, so you know what each one contributes.

Mixing and concentration

The liquid you add sets the concentration. For a 1mg vial:

• 1mL of bacteriostatic water gives 1mg/mL
• 2mL of bacteriostatic water gives 500mcg/mL

Add the water slowly along the inside of the vial and roll it gently until the powder dissolves. Shaking can damage peptide chains.

How to store IGF-1 DES

Keep sealed vials in a freezer at -20°C or a fridge at 2 to 8°C, away from light. After mixing, store the vial in the fridge and use it within about four weeks. Keep nasal spray bottles in the fridge once opened, capped and upright.

Label the container with the opening date. It saves guesswork later.

Working with it safely

Handle it the way you’d handle any research chemical: gloves, eye protection and a clean bench. Don’t eat or drink where you work with it.

Keep it out of reach of anyone who isn’t trained, and dispose of used syringes, vials and wipes according to your lab’s waste rules.
