What is Ibudilast?

Ibudilast is a phosphodiesterase inhibitor sold in Japan for decades as an asthma and post-stroke dizziness drug. MediciNova has developed it as MN-166 for neurological diseases.

How it works

Ibudilast blocks several phosphodiesterases, mainly PDE3, PDE4, PDE10 and PDE11, which raises cAMP. It also blocks macrophage migration inhibitory factor (MIF) and Toll-like receptor 4 signaling on glial cells.

Calming activated glia is the reason it shows up in neuroinflammation and pain research.

Published Ibudilast research

Fox and colleagues reported in the New England Journal of Medicine in 2018 that ibudilast slowed brain shrinkage in progressive multiple sclerosis. It’s also been studied in ALS and in addiction.

Animal work links it to reduced neuroinflammation.

How labs use it

Cognition studies often test a compound in a deficit model, for example after sleep loss, stress or a drug that impairs memory, because effects are easier to see there than in healthy animals.
