Description

About DHEA

DHEA is a steroid made by the adrenal glands. It’s a precursor to testosterone and estrogen.

Levels fall with age.

Mechanism of action

DHEA is converted in tissues into androstenedione and then testosterone or estrogen, depending on the enzymes present. It also acts on its own at some neurosteroid receptors, such as NMDA and sigma-1.

How much becomes testosterone versus estrogen depends on sex and tissue.

Nair and colleagues reported in NEJM (2006) that DHEA didn’t improve body composition in older adults.

It’s on the WADA list.

It’s mostly found in the blood as DHEA sulfate, which acts as a reservoir that tissues convert back to DHEA.

Using DHEA in the lab

Labs studying the HPG axis usually take a baseline hormone panel, then repeat it at fixed points during and after exposure. That makes recovery curves easy to compare between compounds.

Estradiol assays vary a lot in accuracy at low levels, so use a sensitive method if estrogen suppression is the endpoint.

Draw blood at the same time of day for every sample, since testosterone and LH follow a daily rhythm.

Measuring the liquid

At 33mg per mL, every 1mL holds 33mg and every 0.1mL holds 3.3mg. A graduated 1mL dropper or syringe makes those measures accurate.
