What’s in Mass Strong?

4-Andro

4-Andro is 4-androstene-3-beta-ol-17-one, a prohormone that converts to testosterone. It’s closely related to androstenedione.

4-Andro is converted to testosterone by 17-beta-HSD. Because testosterone can aromatize, some of it becomes estradiol.

MK-677

MK-677 (ibutamoren) is an orally active, nonpeptide growth hormone secretagogue developed by Merck. It mimics ghrelin at the GHS-R1a receptor, raising growth hormone and IGF-1. It’s often sold next to SARMs, but it isn’t one.

MK-677 binds GHS-R1a, the same receptor as ghrelin and the GHRP peptides, in the pituitary and hypothalamus. That boosts the size of natural GH pulses and lifts IGF-1 from the liver.

LGD-4033

LGD-4033 (ligandrol, also coded VK5211) is a nonsteroidal SARM discovered by Ligand Pharmaceuticals and later licensed to Viking Therapeutics. It binds the androgen receptor with high affinity and, in animal models, acts strongly on muscle and bone while sparing the prostate.

Like other SARMs, LGD-4033 binds the androgen receptor and the receptor complex moves into the nucleus to switch on androgen-responsive genes. Its binding is unusually tight, with a reported Ki around 1 nanomolar.

Research behind Mass Strong

Research on this family, including studies on androstenedione by King and colleagues in JAMA (1999), found that much of the material turned into estrogen as well as testosterone.

Nass and colleagues ran a two-year trial in healthy older adults, published in the Annals of Internal Medicine in 2008. MK-677 raised GH and IGF-1 to levels seen in young adults and increased fat-free mass, with rises in fasting glucose noted.

A phase 1 study by Basaria and colleagues, published in the Journals of Gerontology in 2013, gave LGD-4033 to 76 healthy young men for 21 days. Lean body mass rose with the dose, and the study also reported dose-related drops in HDL cholesterol and testosterone.
