What is Cardarine?

GW501516 (cardarine) is a PPAR-delta agonist developed by GlaxoSmithKline and Ligand in the 1990s. It’s often sold next to SARMs, but it works through a different receptor, one that switches on fat-burning genes in muscle.

GSK stopped development after rodent studies linked long-term high exposure to cancer in several organs. It’s on the WADA prohibited list.

Mechanism of action

PPAR-delta is a nuclear receptor. When GW501516 binds it, it pairs with the retinoid X receptor and switches on genes for taking up and burning fatty acids, such as CPT1 and PDK4.

In muscle, that shifts fuel use from glucose toward fat and pushes fibers toward the slow-twitch, fatigue-resistant type. That’s the basis for the endurance results in mice.

Cardarine research

Narkar and colleagues showed in Cell in 2008 that GW501516 combined with exercise or with AICAR raised running endurance in mice and shifted muscle toward slow-twitch fibers.

Earlier work by Oliver and colleagues, published in PNAS in 2001, reported higher HDL cholesterol and lower triglycerides in obese rhesus monkeys.
