Description

Our PNC-27 is a p53-derived membrane-active peptide, supplied for lab research. It’s tested to 99% purity or higher and sold strictly for lab research.

PNC-27 explained

PNC-27 is a 32-residue peptide that joins a sequence from p53 (residues 12-26, the HDM-2 binding region) to a membrane-penetrating sequence. It’s studied for its effects on cancer cell membranes.

It’s only been studied in cells and animals.

Mechanism of action

PNC-27’s p53 sequence binds HDM-2, which some cancer cells carry on their outer membrane. Binding there lets the peptide form pores and break the membrane.

Normal cells have little surface HDM-2, which is the proposed reason they’re spared.

PNC-27 research

Sarafraz-Yazdi and colleagues reported in PNAS (2010) that PNC-27 bound HDM-2 on cancer cell membranes and formed pores that killed the cells, while untransformed cells were spared.

Follow-up work looked at leukemia and pancreatic cancer cell lines.

The primary literature is on PubMed. Here’s a search to start from.

Study design notes

Researchers who work with peptides tend to order a small vial first to check solubility and handling, then move to larger sizes once the protocol is settled.

Keep a log of the batch number, the mixing date and the volume of water used. If results drift between runs, those three details are usually where the answer is.

If you’re new to a peptide, read two or three of the original papers before you start. They’ll show which endpoints moved, which didn’t, and how other labs set up their controls.

PNC-27 product details

  • Compound: PNC-27
  • Class: p53-derived membrane-active peptide
  • Size: 5mg
  • Purity: 99% or higher, verified by first- and third-party testing
  • Use: laboratory research only

Quality and testing

We test each batch for identity and purity before it ships, using HPLC and mass spectrometry, with independent third-party checks on top of our own.

The batch number on the label matches the certificate of analysis. If your lab keeps paperwork on file, ask us for it and we’ll email it over.

Storage and shelf life

Keep the container sealed at room temperature, away from heat, moisture and sunlight. Write the date you opened or mixed it on the label, so anyone else in the lab knows how old it is.

Safe handling

Handle it the way you’d handle any research chemical: gloves, eye protection and a clean bench. Don’t eat or drink where you work with it.

Keep it out of reach of anyone who isn’t trained, and dispose of used syringes, vials and wipes according to your lab’s waste rules.

How it compares

DSIP (Delta Sleep-Inducing Peptide): Delta sleep-inducing peptide (DSIP) is a nine-amino-acid peptide first isolated from rabbit blood in 1977 by Schoenenberger and Monnier. It was named for its effect on slow-wave sleep in rabbits.

PT-141: PT-141 (bremelanotide) is a cyclic seven-amino-acid peptide derived from melanotan II. It activates melanocortin receptors, mainly MC4R, in the brain.
